8、OBJECTIVE: To study the comparative gastroprotective effect of Luffa acutangula methanolic extract (LAM) and aqueous extract (LAW) on type II diabetes rats. METHODS: Streptozotocin (65 mg/kg, i.p.) along with nicotinamide (120 mg/kg, i.p.) was used to induce non insulin depen

9、dent diabetes mellitus (NIDDM) in rats. A daily oral dose of aspirin (200 mg/kg, i.p.) was administered for initial seven days to induce gastric ulcerations in the diabetic rats. LAM and LAW were administered orally in the doses of 100, 200 and 400 mg/kg once daily for 21 days. Glibenclamide and ran

10、itidine were used as standards for comparing the antidiabetic and antiulcer effect respectively.RESULTS: LAM significantly (P0.01) increased mucosal glycoprotein and antioxidant enzyme level in gastric mucosa of diabetic rats than LAW (P 0.05). LAM was efficient in reversing the delayed healing of g

11、astric ulcer in diabetic rats close to the normal level. LAM exhibited better ulcer healing effect than glibenclamide and LAW, because of its both antihyperglycemic and mucosal defensive actions.CONCLUSIONS: Thus, LAM is proved to be a better alternative for treating gastric ulcers co-occurring with

Although its gastroprotective activity was reported, the active compounds have not been identified. The

15、refore, the aim of the present study was to identify at least one active compound potentially responsible for the gastroprotective activity of H. suaveolens by using a bioassay guided study with an ethanol-induced gastric ulcer experimental model in rats. The results show that the hexane extract had

16、 protective activity (close to 70% when using doses between 10 and 100 mg/kg), and that the compound suaveolol, isolated from this extract, was one of the active gastroprotective agents. This is the first report about the gastroprotective activity of suaveolol. Rats treated with this compound at 3,

17、10, 30 and 100 mg/kg showed 12.6, 21.3, 39.6 and 70.2% gastroprotection respectively. The effect elicited by suaveolol (at 100 mg/kg) was attenuated by pretreatment with either N(G)-nitro-L-arginine methyl ester (70 mg/kg, i.p.), a nitric oxide (NO) synthase inhibitor, indomethacin (10 mg/kg, s.c.),

18、 a blocker of prostaglandin synthesis, or N-ethylmaleimide (10 mg/kg, s.c.), a blocker of sulfhydryl groups. Ten Wistar rats were used as normal group and fed normal diet. After 24 weeks

24、ght recorded.RESULTS:Similar AMPK mRNA levels were found in the Dan-gua Fang group and normal group, slightly higher than the values obtained for the remaining groups (P0.05). AMPK protein expression in the Dan -gua Fang group animals was similar to other diabetic rats, whereas phospho-AMPK (Thr -17

25、2) protein levels were markedly higher than in the metformin group and simvastatin group (P0.05), respectively. However, phosphor-AMPK /AMPK ratios were similar in all groups. Dan -gua Fang reduced fasting blood glucose with similar strength to metformin, and was superior in reducing cholesterol, tr

26、iglycerides, high-density lipoprotein cholesterol as well as improving low-density lipoprotein cholesterol in comparison with simvastatin and metformin. Dan-gua Fang decreases plasma alanine aminotransferase (ALT) significantly.CONCLUSION:Dan-gua Fang, while treating phlegm-stasis, could decrease BG

27、 and lipid in type 2 diabetic GK rats fed with high-fat diet, and effectively protect liver histomorphology and function. This may be partly explained by increased AMPK expression in liver.
