生物化學I(英)課程號:07110110 周學時:3-0 學分: 3.0英文名稱:Biochemistry (A) I一、教學目的與教學要求:本課程將作為學生全面進行現(xiàn)代生物學其他課程學習的最重要基礎(chǔ)課程,著重開展有關(guān)生物化學基礎(chǔ)(生命的分子理論,細胞,生物分子,水)、分子構(gòu)造和觸媒作用(氨基酸,多肽和蛋白質(zhì),蛋白質(zhì)的三級構(gòu)造,蛋白質(zhì)的功能,酶學,碳水化合物及糖生物學,核苷酸及核酸,脂肪,生物膜及運輸,生物信號)、新陳代謝(生物能學原理,糖酵解,檸檬酸循環(huán),脂肪酸氧化,氨基酸氧化及尿形成,氧化磷酸化及光化磷酸化,碳水化合物的合成,脂肪合成,氨基酸、核酸的合成)及信息通路(基因和染色體,DNA代謝,RNA代謝,蛋白質(zhì)代謝,基因表達的調(diào)控,DNA重組技術(shù))四個專題的講授工作。課程結(jié)合經(jīng)典的和最新的科學研究實例來闡述基本理論和概念,對生化實驗技能具有重要的理論指導意義,并為學生今后從事生命科學相關(guān)的工作打下基礎(chǔ)。二、主要內(nèi)容及學時分配(48學時):1. Biomolecues and Water 3學時Chemical composition and bonding, Three-dimensional structure: configuration and conformation, Chemical reactivity, Macromolecules and their monomeric subunits, Prebiotic evolution, Weak interactions in aqueous systems, Ionization of water, weak acids, and Weak bases, Buffering against pH changes in biological systems, Water as a Reactant, The fitness of the aqueous environment for living organisms.3. Amino Acids, Peptides, and Proteins 6學時Amino acids, Peptides and proteins, Working with proteins, The covalent structure of proteins.4. The Three-Dimentional Structure of Proteins 3學時Overview of protein structure, Protein secondary structure, Protein tertiary and quaternary structures, Protein denaturation and folding.5. Protein Function 3學時Reversible binding of a protein to a ligand: oxygen-binding proteins, Complementary interactions between proteins and ligands: the immune system and immunoglobulins, Protein interactions modulated by chemical energy: actin, myosin, and molecular motors.6. Enzymes 6學時An introduction to enzymes, How enzymes work, Enzyme kinetics as an approach to understanding mechanism, Examples of enzymatic reactions, Regulatory enzymes.7. Lipids 3學時Storage Lipids, Structural Lipids in Membranes, Lipids as Signals, Cofactors, and Pigments, Separation and Analysis of Lipids.8. Biological Membranes and Transport 3學時The molecular constituents of membranes, The supramolecular architecture of membranes, Solute transport across membranes.9. Biosignaling, Structure and Function of Hormones 3學時Molecular mechanisms of signal transduction, Gated ion channels, Receptor enzymes, G protein-coupled receptors and second messengers, Sensory transduction in vision, olfaction, and gestation, Structure and function of hormones, Regulation of hormone., Regulation of transcription by steroid hormones, Regulation of the cell cycle by protein kinases, Oncogenes, tumor suppressor genes, and programmed cell death. 10. Nucleotides and Nucleic Acids 3學時Some basics, Nucleic acid structure, Nucleic acid chemistry, Other functions of nucleotides.11. Genes and Chromosomes 3學時Chromosomal Elements, The Size and Sequence Structure of DNA Molecules, DNA Supercoiling, Chromatin and Nucleoid Structure.12. DNA Metabolism 6學時A Word about Terminology, DNA Replication, DNA Repair, DNA Recombination.13.
