Magnesium(Mg)deficiencymayplayanimportantroleinpathogenesisofenhancedvascularreactivityinhypertension.TheoverallhypothesistobeevaluatedisthatMgdeficiencycausedbyglucoseintolerance,insulinresistanceorotherfactorsinhypertensivesleadstoincreasedvasomotortoneviaalteredreleaseofvasoactivecyclooxygenaseandlipoxygenaseproductsofarachidonicacidandenhancedangiotensinII(AII)action.ToevaluatetheeffectsofMgdeficiencyinnormalsubjects,wewillinducetheconditionbyadministrationofalowMgdiet.VascularandadrenalsensitivitytoAII,plateletaggregationandeicosanoidlevelswillbestudiedpriortoandafterMgdeficiencyisestablished.SinceevidencesuggestsMgdeficiencycanmodulateinsulinaction,theeffectofthisdeficiencyonglucosetolerancewillalsobestudied.InanotherprojecttheeffectofinsulinonintracellularMglevelswillbestudiedusinganewfura2Mgdyetechnique.Thesestudieswillbeperformedingroupsofsubjectswithvariedbloodpressureandinsulinlevels.AlsotheeffectsofacuteintravenousandchronicoralMgloadingontheaboveparameterswillbestudiedinsimilarsubjectgroups.WewilldirectlystudytheeffectofMgonAII,insulinandinsulinlikegrowthfactoractioninisolatedandculturedadrenalglomerulosacells.ConcentrationofMgwillbevariedandsignaltransductionandsteroidogeniceffectswillbeevaluate.Thesestudieswillprovideinsightintomechamismsimportanttothepathogenesisofalteredvascularreactivityofsubjectswithhypertensionorhyperinsulinemia.

SpecificAims:(1)

DeterminetheeffectsoflowMgonvascularandadrenalsensitivitytoAII(plateletaggregationandeicosanoidlevels,glucosetolerance);(2)determinetheeffectofinsulinonintraceullarMglevels(fura2Mgdyetechnique).Thesestudieswillbeperformedingroupsofsubjectswithvariedbloodpressureandinsulinlevels;(3)determinetheeffectofacutei.v.andchronicoralMgloadingontheaboveparameters;(4)determinethesignaltransductionandsteroidogeniceffectsofMgandAII,insulinandinsulin-likegrowthfactoractioninisolatedandculturedadrenalglomerulosacells.

Significance:Thesestudieswillprovideinsightintomechamismsimportanttothepathogenesisofalteredvascularreactivityofsubjectswithhypertensionorhyperinsulinemia.
