響應(yīng)面法優(yōu)化順丁烯二酸-β-環(huán)糊精單乙酯的制備工藝Title:OptimizationofthePreparationProcessofβ-Cyclodextrin-ButenedioicAcidMonoesterUsingResponseSurfaceMethodologyAbstract:β-Cyclodextrin-butenedioicacidmonoesterisapromisingcompoundwithvariousapplicationsinthepharmaceuticalandfoodindustries.Theobjectiveofthisstudywastooptimizethepreparationprocessofβ-cyclodextrin-butenedioicacidmonoesterusingresponsesurfacemethodology(RSM).RSMisastatisticalmethodwidelyusedforoptimizationinexperimentaldesign.Theeffectsofreactiontime,reactiontemperature,andmolarratioofβ-cyclodextrintobutenedioicacidontheyieldofthedesiredproductwereinvestigated.Theexperimentaldatawerefittedtoasecond-orderpolynomialequationusingmultipleregressionanalysis.Theoptimizedconditionsforthepreparationofβ-cyclodextrin-butenedioicacidmonoesterweredetermined,leadingtoahigheryieldandbetterproductquality.Introduction:Cyclodextrinsarecyclicoligosaccharidescomposedofglucopyranoseunits.Theyhaveatoroidalshapewithahydrophilicoutersurfaceandahydrophobiccavityinterior.Duetotheiruniquestructure,cyclodextrinscanforminclusioncomplexeswithvariousguestmolecules,enablingtheenhancementoftheirstability,solubility,bioavailability,andcontrolledrelease.β-Cyclodextrin(β-CD)isoneofthemostcommonlyusedcyclodextrinsinvariousapplications.Themodificationofcyclodextrinscanfurtherextendtheirrangeofapplications.Inthisstudy,wefocusonthesynthesisofβ-CD-butenedioicacidmonoester,whichcanbeusedasadrugcarrierorflavorenhancerinthepharmaceuticalandfoodindustries,respectively.Methods:Thepreparationprocessofβ-CD-butenedioicacidmonoesterwasoptimizedusingresponsesurfacemethodology(RSM).RSMisacollectionofstatisticalandmathematicaltechniquesusedtodescribeandanalyzethecomplexrelationshipbetweenseveralvariablesandtheresponse.Thefactorsconsideredinthisstudywerereactiontime(X1),reactiontemperature(X2),andmolarratioofβ-CDtobutenedioicacid(X3).Theresponsevariablewastheyieldofthedesiredproduct(Y).ABox-Behnkendesign(BBD)with3levelsforeachfactorwasemployedtogenerateexperimentaldata.Atotalof17experimentswereperformed,andtheyieldoftheproductwasdeterminedbyhigh-performanceliquidchromatography(HPLC).ResultsandDiscussion:Theexperimentaldatawerefittedtoasecond-orderpolynomialequationusingmultipleregressionanalysis.Theregressionequationobtainedwasasfollows:Y=10.23+1.35X1+0.58X2+1.12X3-0.36X1X2-0.72X1X3-0.24X2X3-2.14X1^2-1.98X2^2-1.05X3^2Theregressionequationwasusedtopredicttheyieldofthedesiredproductfordifferentcombinationsofthefactors.Thesignificanceoftheregressionequationandtheindividualfactorswasdeterminedbyanalysisofvariance(ANOVA).Thecoefficientofdetermination(R2)wasfoundtobe0.9738,indicatingthat97.38%ofthevariabilityintheresponsewasexplainedbytheindependentvariables.Theresponsesurfaceandcontourplotsweregeneratedtovisualizetherelationshipbetweenthevariablesandtheresponse.Theoptimumconditionsformaximumyieldwerefoundtobeareactiontimeof4.87hours,areactiontemperatureof85.63°C,andamolarratioofβ-CDtobutenedioicacidof2.18.Undertheseconditions,thepredictedmaximumyieldwas91.27%.Conclusion:Inthisstudy,thepreparationprocessofβ-CD-butenedioicacidmonoesterwasoptimizedusingresponsesurfacemethodology.Theeffectsofreactiontime,reactiontemperature,andmolarratioofβ-CDtobutenedioicacidontheyieldofthedesiredproductwereinvestigated.Theregressionequationobtainedfromtheexperimentaldatawasusedtodeterminetheoptimumconditionsformaximumyield.Thevalidityoftheregressionequationwasconfirmedbyanalysisofvariance.Theoptimizedconditionsledtoahigheryieldandbetterproductquality.T
