1、頸痛寧丸論文:頸痛寧丸對(duì)椎動(dòng)脈型頸椎病模型家兔治療作用的研究【中文摘要】探討頸痛寧丸對(duì)椎動(dòng)脈型頸椎病治療作用及相關(guān)機(jī)制。方法:取健康新西蘭大耳白家兔70只,雌雄各半,體重2.5-3.0kg,各家兔編號(hào)。采用TCD檢測(cè)各家兔腦血流,記錄各家兔椎-基底動(dòng)脈血流速度。采用單側(cè)頸椎C3-C5橫突側(cè)面注射硬化劑法制備椎動(dòng)脈型頸椎病家兔模型。再次經(jīng)TCD檢測(cè)椎-基底動(dòng)脈血流速度,確定模型制備成功。挑選40只成功模型家兔,隨機(jī)分為5組,每組8只。陽(yáng)性對(duì)照組給予頸復(fù)康顆粒1.0g/kg、頸痛寧丸高、中、低劑量組分別給予頸痛寧丸2.0g/kg、1.0g/kg、0.5g/kg,模型組給予等容積生理鹽水,另取8只未

2、造模家兔作為空白對(duì)照組,動(dòng)物正常飼養(yǎng)。各組動(dòng)物每日給藥一次,連續(xù)4周,于給藥第27天進(jìn)行TCD檢測(cè),第28天藥后30min采血、取頸部注射部位肌肉組織和腦組織。以化學(xué)法檢測(cè)家兔血清NO濃度;酶聯(lián)免疫法檢測(cè)血漿ET含量、IL-1含量、TNF-a含量;在光鏡下觀察頸部注射部位肌肉和腦組織形態(tài)、細(xì)胞結(jié)構(gòu)、結(jié)締組織、炎性因子、瘀血等情況。另取小白鼠50只,隨機(jī)分為5組,空白對(duì)照組正常飼養(yǎng),陽(yáng)性對(duì)照組給予阿司匹林0.3 g/kg,頸痛寧丸高、中、低劑量組分別給予頸痛寧丸5.4g/kg.2.7g/kg.1.35g/kg.各組動(dòng)物每日給藥一次,連續(xù)7天。末次藥后30min,通過乙酸刺激扭體反應(yīng)觀察頸痛寧丸的

3、鎮(zhèn)痛作用。結(jié)果:1.頸痛寧丸對(duì)椎動(dòng)脈型頸椎病模型家兔治療作用的研究:TCD檢測(cè)結(jié)果表明,給藥后頸痛寧丸高、中、低劑量組左椎動(dòng)脈Vm、右椎動(dòng)脈Vm、基底動(dòng)脈Vm與給藥前相應(yīng)速度比有顯著增加。其中高劑量組給藥后左椎動(dòng)脈Vm、右椎動(dòng)脈Vm、基底動(dòng)脈Vm與給藥前相應(yīng)速度比有顯著差異(P<0.01、P<0.01、P<0.05),中劑量組給藥后左椎動(dòng)脈Vm、右椎動(dòng)脈Vm與給藥前相應(yīng)速度比有顯著差異(P<0.05)。用藥后,各給藥組與模型對(duì)照組比,左側(cè)椎動(dòng)脈Vm、右側(cè)椎動(dòng)脈Vm、基底動(dòng)脈Vm均有不同程度的提高。其中高劑量組左側(cè)椎動(dòng)脈Vm、右側(cè)椎動(dòng)脈Vm、基底動(dòng)脈Vm與模型對(duì)照組比有明

4、顯差異(P<0.01、P<0.05、P<0.01),中劑量組左側(cè)椎動(dòng)脈Vm與模型對(duì)照組比有明顯差異(P<0.05)。血液檢測(cè)結(jié)果顯示,頸痛寧丸高、中劑量組血漿中ET含量與模型對(duì)照組比較有顯著減少(P<0.01、P<0.05);頸痛寧丸高、中劑量組血漿IL-1含量與模型對(duì)照組比較顯著減少(P<0.05);頸痛寧丸高、中劑量組血漿TNF.含量與模型對(duì)照組比明顯降低(P<0.01、P<0.05);頸痛寧丸高、中劑量組血清NO含量與模型對(duì)照組比明顯升高(P<0.01、P<0.05)。光鏡下觀察得出,各給藥組頸部注射部位肌肉組織和腦組織細(xì)

5、胞結(jié)構(gòu)變化和結(jié)締組織增生、炎性細(xì)胞數(shù)量、瘀血等現(xiàn)象較模型對(duì)照組比有明顯改善。2.頸痛寧丸抗痛作用的實(shí)驗(yàn)研究:各給藥組動(dòng)物扭體次數(shù)較空白組比明顯減少,其中頸痛寧丸高、中劑量組扭體次數(shù)與模型對(duì)照組比有顯著差異(P<0.05)。結(jié)論:頸痛寧丸對(duì)椎動(dòng)脈型頸椎病家兔有一定的治療作用,其作用機(jī)制可能與改善血液循環(huán),調(diào)節(jié)血管舒縮,抑制炎性因子,增強(qiáng)機(jī)體抗痛能力有關(guān)。【英文摘要】:To explore the effects and mechinisms of Jingtongning-pill (JTNP), which has effects on cervical spondylotic vert

6、ebral arteriopathy(CSA).Methods:70 healthy New Zealander rabbits in half respectively male and female,weighing 2.5kg to 3.Okg,were numbered.All rabbits were measured the mean velocity (Vm) in vertebral arteries(VA) and basilar arteries(BA) using transcranial doppler(TCD).Then all rabbits were inject

7、ed tissue sclerosing liquid into the soft tissue on the left side of cervical vertebral transverse protuberance to set up the CSA models.Measured the Vm in left VA,right VA and BA using TCD once more to confirm the successful models.Next,Selected 40 successful model rabbits and grouped them into 5 g

8、roups randomly,8 in each group.All groups were administered of relevant drugs for 4 weeks,once a day.The positive group was administered Jingfukang-granule1.0g/kg.JTNP was administrated to the high-dose group 2g/kg,the middle-dose group 1g/kg,the low-dose group 0.5g/kg,respectively. The model contro

9、l group was given same amount of saline. The normal group was composed of another 8 healthy rabbits and was fed on normal diet.27days after first administration,all rabbits were measured the mean velocities in left VA,right VA and BA using TCD.Next day,tested the content of ET,IL-1,TNF-in plasma,NO

10、in serum,and observed the histopathological change of the injection tissue and brain by light microscope about histomorphology,cytoarchitecture,hyperplasia of connective tissue, increaseing of inflammatory cells,blood stagnation and so on.Besides,50 mouses were randomly signed into 5 groups,10 each

11、group.The positive control group was administered of aspirin0.3g/kg.JTNP was administered to the high-dose group 5.4g/kg,the middle-dose group 2.7g/kg,the low-dose group 1.35g/kg,respectively.The normal control group was given same amount of saline,all groups were administered for a week.Half an hou

12、r after the last administration,all groups were injected acetic acid into belly and then observed the amount of body twisting in 15 minutes.Results:1.The detecting results by TCD show that after administration the mean velocities in left VA, right VA and BA were faster than the mean veloci-ties befo

13、re administration.The mean velocities in left VA, right VA and BA of the high-dose group were very different from the mean velocities in left VA, right VA and BA of the model control group(P<0.01,P<0.01,P0.05,P<0.01).The mean velocities in left VA and right VA of the middle-dose group were

14、also different from the mean velocities before administration(P<0.05).After administration, the mean velocities in left VA, right VA and BA were faster than the mean velocities in left VA and right VA of the model control group,the mean velocities in left VA, right VA and BA of the high-dose grou

15、p were very different from the mean velocities in left VA and right VA of the model control group(P<0.01,P<0.05,P<0.01).The Vm in left VA of the middle-dose group was also different from the Vm in left VA of the model control group(P<0.05).In the high-dose group and the middle-dose group

16、,the content of NO in serum was obvious higher than the model control group(P<0.01,P<0.05).In the high-dose group and the middle-dose group,the ET content in plasma was obvious lower than the model control group(?<0.01,P<0.05).In the high-dose group and the middle-dose group,the content

17、of IL-lin plasma was obvious lower than the model control group (P<0.05). In the high-dose group and the middle-dose group,the content of TNF-a in plasma was obvious lower than the model control group(P<0.01,P<0.05).By light microscope the observed results showed that the phenomenons,such a

18、s histomorphology,Cytoarchitecture, hyperplasia of connective tissue,increasing of inflammatory cells,blood stagnation and so on,of all tested groups were superior to the model control group. 2.JTNP analgesia study showed that the times of body twisting of the high-dose group and the middle-dose gro
